(PR040) Evaluation of biologics FRIL and PG1 in Endodontic Treatment: A Targeted Approach Against Biofilm-Forming Pathogens

Objective: This study evaluates the potential of biologics PG1 and FRIL as alternatives to calcium hydroxide Ca(OH)2 for interim treatment in endodontic infections, targeting biofilm integrity in necrotic teeth. Ca(OH)2 lacks specificity against biofilm-forming pathogens, leading to the hypothesis that the combined action of PG1, an antimicrobial peptide, and FRIL, a lectin binding bacterial glycoproteins, will provide superior antibacterial efficacy against aerobic (S. mutans) and anaerobic (P. gingivalis, and F. Nucleatum) bacteria.

Methods: Endodontic microbial samples from necrotic teeth with apical periodontitis were divided into a control group grown on blood agar without treatment and a study group treated with 100 µg/mL PG1 and 25% concentrated FRIL extract for 1 hour at 37°C, under anaerobic conditions. Sample size ranged from 5-10 per group. Each sample (10µL, treated and untreated) was inoculated onto blood agar plates and selective media plates and incubated for five days. Total viable colonies (CFU/mL) were counted on day 5 of incubation.

Results: The study group treated with PG1 and FRIL showed no growth of anaerobic (P. gingivalis, and F. Nucleatum) bacteria post-incubation for 5 days at 37°C, indicating effective biofilm disruption and bacterial eradication. Significant bacterial growth was observed in the control group, with results confirming the enhanced antimicrobial efficacy of the biologic agents.

Conclusion: The combination of FRIL and PG1 effectively disrupted biofilm and eliminated anaerobic bacterial growth suggesting their potential as more targeted and effective alternatives to Ca(OH)2 in treating necrotic teeth. Future studies with larger sample sizes are recommended to further support these findings.
